By using a dedicated culture container (microtranswell) for this device, it is possible to allow the culture medium to permeate into the interior of the cultured tissue or cell layer, similar to blood flow. 【Features】 ■ By conducting cell culture in an environment close to in vivo conditions with this device, the following outcomes are expected: (1) The culture medium can be supplied to deeper parts of the tissue or cell mass, enabling longer culture periods and maintenance of activity. (2) Shear stress from flow allows observation of the mechanical responses of cells, such as angiogenesis, which are difficult to observe in static cultures. (3) Secreted substances from upstream cells can affect downstream cells, enabling sequential reactions. ■ The delivery pressure can be adjusted with a controller. ■ The culture system has a sealed structure, allowing for culture in a horizontal position, so the entire holder can be placed on a microscope stage, enabling observation of the culture area from the side of the culture container (see conceptual diagram). ■ The entire system is compact, allowing it to be installed within your existing incubator, making it easy to try perfusion culture. * When using scaffold materials, it is necessary to select a type that allows for easy permeation of the culture medium. 【Applications】 - Cultivation Results [1] HepG2 cells (human liver cancer-derived cell line) were seeded, and the expression levels of CYP genes were evaluated 20 days after cultivation.
