Unlike mice and rats, hamsters exhibit human-like cholesterol and bile acid metabolism characteristics. Therefore, pharmacological tests with the FXR agonist obeticholic acid show human-like weight and liver inflammation suppression, as well as side effects such as increased LDL cholesterol and decreased HDL cholesterol. (In mice and rats, this cholesterol side effect is reversed, with decreased LDL cholesterol and increased HDL cholesterol.) Physiogenex has adopted a cafeteria diet approach to induce this hamster model for overfeeding and NASH (non-alcoholic steatohepatitis) by providing a high-fat diet and high fructose water. This induces obesity, insulin resistance, hypercholesterolemia, and elevated ALT levels.
