■Structure of Nucleic Acids Nucleic acids refer to deoxyribonucleic acid (DNA) and ribonucleic acid (RNA). They have a repetitive structure of nucleotides. The building blocks, nucleotides, consist of a nucleic acid base, a five-carbon sugar (ribose), and phosphate. ■Nucleic Acid Medicines Nucleic acid medicines are gaining attention as a new drug discovery modality. They are drugs based on natural or chemically modified nucleotides, which act directly on the body without mediating gene expression and are characterized by being produced through chemical synthesis. ■Synthesis of Nucleic Acids (Oligonucleotides) Current nucleic acid (oligonucleotide) synthesis primarily uses the solid-phase synthesis method (phosphoramidite method) developed in the 1980s. Nucleotides are added on solid-phase supports such as polymer beads, repeating the synthesis cycle until the desired chain length is achieved, with the following steps 1 to 4 constituting one cycle. 【Synthesis Cycle】 1.
