HexAuFoil samples support improves every image! CryoEM inherently has issues with data loss caused by particle movement during imaging, which leads to a decrease in mapping quality. While data processing can mitigate this data loss, advanced sample processing techniques are required. To address these issues, we propose a solution using the new cryoEM grid "HexAuFoil," which has been launched by Quantifoil, a group company of SPT Labtech.

HexAuFoil is composed of gold foil with holes arranged on a hexagonal gold mesh grid. The hexagons of the grid are approximately 50 μm in size, and the outer shell diameter is about 10-12 μm. The gold foil has holes with a diameter of 290 nm arranged in a hexagonal pattern, with a spacing of 310 nm between the holes, forming a repeating structure of 600 nm. The ultra-small pore gold support reduces particle movement to below the angstrom level. The small pore size provides additional stability to the thin ice layer supporting the sample, reducing or eliminating ice warping due to cryo-cooling. The position of the particles stabilizes, allowing for mapping to be estimated to zero electron loss before radiation damage occurs. Efforts are being made to reduce particle movement while improving the throughput of automated cryoEM without compromising map quality, incorporating features such as maximum pore density.
