Case Studies on the Analysis of Excipients Using HPLC (English Material)

Analysis of Pharmaceutical Excipients – What is Universal Chromatography Analysis for Complex Mixture Analysis?

Excipients are substances added for the purpose of shaping, bulking, and diluting solid formulations, and they consist of a mixture of compounds with various chemical properties. Both excipients and active ingredients need to be managed appropriately, and profiling their impurities is also important. What analytical methods are suitable for the detection and quantification of a wide range of excipients? Check out our ebook that covers analytical examples solved by Charged Aerosol Detectors (CAD). You are sure to find the excipient analysis you are looking for. This ebook (in English) includes the following information: • Excipients applicable to pharmaceutical manufacturing and their uses • What columns are effective for excipient analysis? Selection considering sample amount and hydrophobicity • How to detect excipients using Charged Aerosol Detectors (CAD) • Examples of excipient analysis in formulations using CAD and information from scientific papers

Overview of the Acclaim column

A column that meets high-difficulty separation needs.

Thermo Scientific Acclaim columns are ideal for high-efficiency high-performance liquid chromatography (HPLC) separations. Their unique features enable challenging and complex separations, allowing for the separation of compounds that traditional HPLC columns cannot achieve. The Thermo Scientific Acclaim columns utilize standard surface chemistry features, including standard functionalities such as 120 C18, C8, Phenyl, Polar Advantage, polar embedded phases, HILIC, as well as mixed-mode functionalities (Thermo Scientific Acclaim Trinity P1 and P2 columns), and application-specific phases (such as organic acids, surfactants, pesticides, contaminants, etc.), along with specialized columns like size exclusion columns (SEC). Furthermore, some Acclaim columns are optimized to operate under the ultra-high-performance liquid chromatography (UHPLC) requirements of the Thermo Scientific Vanquish UHPLC system.

AAV capsid analysis column

ProPac columns support virus vector analysis.

Adeno-associated virus (AAV) is a suitable vector for many gene therapies. The Empty/Full capsid ratio of AAV is an extremely important quality characteristic that needs to be monitored throughout the entire development cycle. The Thermo Scientific ProPac column series leverages approximately 20 years of experience in ion exchange to continue supporting researchers in the biotherapeutics field. The Thermo Scientific ProPac SAX-10 LC column easily separates the Empty/Full capsid bodies of AAV serotype 6 and AAV serotype 8 in less than 10 minutes, allowing for immediate quantification results with minimal sample pretreatment.

Acclaim Mixed-Mode HPLC Column

A column that is expected to provide high separation.

Liquid chromatography aims to achieve good separation ability to obtain meaningful information through peak resolution. In mixed-mode chromatography, numerous parameters can be adjusted to optimize separation efficiency, allowing for effective control of separation performance. The Thermo Scientific Acclaim mixed-mode column features unique surface chemistry that facilitates easy adjustment of analyte elution, enabling maximum separation efficiency, which often leads to successful separation of compounds that cannot be addressed by other conventional high-performance liquid chromatography (HPLC) columns. The combination of ion exchange and reversed-phase, specifically HILIC chemistry, can extract the chemical properties of sample mixtures, allowing for complete separation of components in complex samples or high-sensitivity detection of trace samples. However, it is impossible to retain highly polar or charged analytes using only reversed-phase chromatography. Ion exchange (IEX) chromatography alone is insufficient for separating hydrophobic compounds. By using columns that combine different functionalities, a completely new toolkit is obtained for separating and evaluating samples.

Options for protein preparation separation: Presentation of technical documentation for HIC columns.

Please obtain the materials regarding the detection of impurities in monoclonal antibodies using hydrophobic interaction chromatography from "PDF Download."

In recent years, there has been an increasing demand for hydrophobic interaction chromatography (HIC) columns for the analysis of protein formulations. Unlike reverse phase liquid chromatography, which denatures proteins and effectively destroys their original structure, HIC preserves the native structure and biological activity of proteins, making it useful for downstream functional analyses such as binding and cell-level potency assays. The MAbPac HIC-10 LC column and MAbPac HIC-20 column are HIC columns with wide-pore silica particles featuring patented alkylamide groups, suitable for the separation of oxidized and deamidated variants of protein formulations. The MAbPac HIC-Butyl column is an HIC column that binds butyl groups to a hydrophilic polymer matrix at an appropriate density, which helps reduce sample adsorption to the filler and suppress carryover, making it suitable for the analysis of antibody-drug conjugates (ADCs). You can view examples of analyses using our HIC columns by downloading the PDF.

[Case Study] AAV Titer Measurement After Downstream Process

By adopting size exclusion chromatography and highly sensitive fluorescence detection, accurate and reproducible measurements can be achieved with a small amount of sample!

We introduce a rapid and cost-effective method for measuring the titer of adeno-associated virus (AAV) products after downstream processing, combining size exclusion chromatography (SEC) with a highly sensitive fluorescence detector. By using the Thermo Scientific MAbPac SEC-1 column, efficient separation of AAV capsids and low molecular weight impurities is achieved, allowing for detailed monitoring of product quality attributes (PQA). This method enables rapid determination of AAV product titer after a single step of affinity purification, contributing to the evaluation of product characteristics. 【Features】 ■ Rapid and cost-effective titer measurement method ■ Measurement possible with small sample sizes due to high-sensitivity fluorescence detection ■ Efficient separation using the MAbPac SEC-1 column ■ Detailed monitoring of PQA is possible ■ Improved reproducibility with low-adsorption glass vials (RSD less than 5%) *For more details, please download the PDF or feel free to contact us.
