Fluorescence has become a major challenge in Raman spectroscopy. Timegate Instruments' patented Timegated Raman technology was developed to address this challenge. The phenomena of Raman scattering and fluorescence occur on different time scales. Raman scattering occurs on a very short sub-picosecond (less than one trillionth of a second) time scale, while the decay time of fluorescence is characterized by being very long. By using time-gate technology, it not only distinguishes between fluorescence and Raman signals but also provides temporal information about the Raman signals. Additionally, delayed noise components such as thermal radiation are ignored in Timegate Raman, which means they do not affect measurements. This allows for the analysis of structural changes in materials that undergo polymorphism at high temperatures (500-900°C), such as spodumene, while suppressing the effects of fluorescence. Free membership registrationWe would like to introduce a case study by Proteon Pharmaceuticals, a Polish company, which evaluated the amplification efficiency of Salmonella enterica enteritidis bacteriophage using the CellMaker single-use bioreactor system. The optical density of the bacteria was measured every 30 minutes. The dynamics of the optical density (OD) were compared between the culture of the bacteria alone and the culture with the addition of phage. The test results demonstrated successful culture and high-efficiency amplification of Salmonella enterica enteritidis serotype. Furthermore, process optimization was conducted to improve phage yield, and this process is now adopted as part of the production process. First, 5L of low-sodium LB broth was pumped into the CellMaker bioreactor bag, and Agrobacterium inoculum was added. The optical density (OD) was measured at 600nm, and the initial OD value was recorded as 0.05. The temperature was maintained at 25°C by a Peltier plate mounted on the back of the enclosure unit. The experiment was stopped the following morning, approximately 15 hours after infection by the phages, and the final concentration of the elution was 1.25x10^10 PFU/mL.
