FabRICATOR (IdeS) is a unique enzyme that digests specific sites just below the hinge region of antibodies. It generates a uniform pool of F(ab')2 and Fc/2 with a 30-minute incubation. No substrates other than IgG are known. Typically, IgG is digested within 30 minutes, but there is no risk of excessive digestion even with longer incubation times. FabRICATOR digests IgG under physiological conditions, maintaining its immunoreactivity. The enzyme activity is maximized at 37°C, but it can also be used at room temperature by extending the reaction time. FabRICATOR digests all subclasses of IgG from humans, monkeys, rabbits, and sheep. However, its activity against mouse IgG2a and IgG3 is limited.
