Liposomes have grown to become an important biomedical carrier system. One of the most significant factors affecting the performance of liposomes is particle size. Methods to reduce the size of liposomes include high-pressure homogenization, extrusion, and ultrasonic irradiation. Among these, extrusion is a relatively mild and rapid method. The Genizer jacketed liposome extruder uses a nitrogen gas cylinder to extrude a liposome suspension of drugs, proteins, or genes through a track-etched membrane with a specified pore size. Typically, about 5 to 10 passes are sufficient to create samples of a consistent liposome size. The Genizer jacketed liposome extruder is designed to minimize dead volume and produce uniform unilamellar liposomes or lipopolyplexes.

The operating procedure is easy and simple. First, set the support disk with a large hole onto the filter support base, then place the laser disk (with the smooth side facing up), one drain disk, and finally the liposome membrane (with the glossy side facing up) in that order. A high-pressure nitrogen tube is standard equipment. By connecting this to the gas cylinder regulator, the GJE-100mL is complete. To generate pressure, simply set the sample in the thermal barrel and turn on the switch of the regulator. To achieve nanoparticle sizes, the sample needs to pass through the track-etched nanopore membrane.
