Three-dimensional cell culture substrate "HYDROX"
66025/66026 Three-Dimensional Nanofiber "HYDROX" Powder Regular/Large
The new type of three-dimensional cell culture substrate "HYDROX" allows for three-dimensional culture of various types of cells simply by seeding a cell suspension.
Features of HYDROX - Derived from chemically synthesized polymers - Ready-to-use, can be stored at room temperature - Forms spheroids simply by seeding cells - Easy recovery of cells after culture - Confirmed usefulness data for culturing liver and nerve cells Currently, in three-dimensional culture, animal-derived components are primarily used as scaffolding materials to promote interactions between cells and between cells and the extracellular matrix. However, the use of animal-derived components has raised issues regarding reproducibility, safety, and handling difficulties. Shimadzu Corporation's three-dimensional nanofiber "HYDROX" is a new type of three-dimensional cell culture substrate using polysarcosine-polylactic acid polymer. It enables three-dimensional culture of various types of cells simply by seeding a cell suspension.

basic information
HYDROX is a powdered reagent made from amphiphilic polymers consisting of polylactic acid and polysarcosine, without using any animal-derived components. The HYDROX polymer powder dissolved in ethanol is dropped onto a culture plate and dried to form a dry gel on the bottom surface of the plate. 1) Add a cell suspension to the created gel. 2) The dried gel absorbs moisture and forms three-dimensional nanofibers called "HYDROX." 3) During this process, the cells trapped in the nanofibers aggregate, forming a three-dimensional cell mass. 4) By adding an excess amount of moisture, the structure of the HYDROX nanofibers is disrupted, allowing for easy recovery of the cell mass alone.

Applications/Examples of results
Immature liver cells induced to differentiate for 14 days from human iPS cells were cultured three-dimensionally using HYDROX. The immature liver cells cultured with HYDROX aggregated over time to form cell clusters (spheroids).
