Industry-first high-throughput gel filtration PhyTip columns for desalting and buffer exchange at small scale and multiple samples.
The chip-type gel filtration PhyTip column filled with size exclusion chromatography resin is optimal for desalting protein samples and exchanging buffer, and it also enables high-throughput screening (HTS) in a 96-well plate. Desalting using the gel filtration PhyTip column can be set up as a secondary purification step after affinity purification with an affinity PhyTip column, such as the removal of imidazole after IMAC affinity purification, using the same automated protocol. Like other PhyTip columns, the resin is packed between two hydrophilic screens; however, unlike other PhyTip columns, the gel filtration column passes the sample from the top. The gel filtration PhyTip column has size exclusion resins of 5K and 30K, which retain low molecular weight compounds while allowing molecules of 5 kDa and 30 kDa or larger to pass through. Unlike other PhyTip columns, the use of the ME system is not required.
