The substantial difference in emission profiles between bound and unbound dyes underlies the "light-on" mechanism.

■Water-soluble dye Acoerela dye is highly water-soluble and does not cause false positives that have been problematic in flow cytometry analysis of extracellular vesicles. Unlike the PKH26, DiR, and DiD dye families, it does not form micelles or nanoparticles when reconstituted in aqueous buffers.
