**Abstract** In recent years, with the high integration of various devices, evaluation in localized areas has become extremely important for product development and functional enhancement. The newly introduced RBS device at the Toray Research Center is equipped with a microbeam line capable of converging high-speed ion beams to a minimum of 1 μm in diameter, enabling precise composition and density analysis in small areas. Additionally, the increase in incident ion species and the acceleration of incident ions have significantly improved the sensitivity for light elements and the mass resolution for heavy elements compared to conventional methods. The high-frequency heating type temperature-programmed desorption analysis device IH-TDS1700 is an analytical instrument that allows real-time observation of molecules desorbing from a sample by electromagnetic induction heating in ultra-high vacuum. Due to the ultra-high vacuum, it enables low background and high-sensitivity measurements. The influence of side reactions caused by atmospheric or generated gases is also significantly suppressed, allowing for the observation of desorbed molecules from the sample under ideal conditions. For metal samples with good heating efficiency, heating above 1700°C is possible. Additionally, low-temperature TDS measurements of diffusible hydrogen in steel, which is a factor in hydrogen embrittlement of steel materials, can also be performed. The control of the main body of the device is automatically managed by a PLC (Programmable Logic Controller), and a touch panel is used for the interface.

[Analysis Case] Characterization of Glycosylated Proteins (LC-TOFMS)

It is possible to efficiently advance the development and quality assurance of biopharmaceuticals!

The LC-TOFMS (Liquid Chromatography Time-of-Flight Mass Spectrometry) method is useful for the characterization analysis of proteins (such as molecular weight analysis, peptide mapping, and glycan analysis). We will introduce a case study of the analysis and examination of the glycan (glycoform and glycan attachment sites) of IgG antibodies using the LC-TOFMS method. ■Analysis Sample: IgG Antibody ■Analysis Method: LC-TOFMS ■Analysis Results - Glycoform Analysis By analyzing the molecular weight, it is possible to estimate the glycoforms of glycoproteins. LC-TOFMS measurement of the IgG antibody revealed the presence of five different glycoforms. - Glycan Attachment Site Analysis The IgG antibody was fragmented through enzymatic treatment and measured by LC-TOFMS. The analysis of the measurement results using dedicated software showed that a N-linked glycan (G0F, G1F, G2F) was attached to the asparagine of the peptide fragment with the amino acid sequence EEQFNSTFR. *For detailed results, please refer to the PDF document.
