High-sensitivity measurement of harmful metals in environmental samples

Introduction of analysis examples using iCAP PRO XP ICP-OES.

Using the Thermo Scientific iCAP PRO XP Duo ICP-OES, we directly analyzed certified reference materials for representative river water and tap water in the environmental field, and we will introduce the results obtained and the performance of this device. In the measurement of certified reference materials for river water and tap water, we demonstrated excellent detection limits and quantitative accuracy through direct introduction. The device's introduction system utilized a general aqueous solution introduction system, and the plasma conditions were measured using Thermo Scientific Qtegra ISDS software. The absolute calibration method was adopted, and the validity of the analytical method was proven through the results of spiking recovery tests. Additionally, the eUV mode, specialized for the ultraviolet region, enables highly sensitive measurements of elements such as As, Cd, Pb, Sb, and Se. Furthermore, by measuring the high-concentration elements, which are the main components, using lateral photometry, we were able to measure over a wide dynamic range without dilution while avoiding ionization interference. Quality control of oligonucleotides requires confirmation of mass, determination of sample purity, and yield. Oligonucleotides show strong absorption at 260 nm, making quantification easily achievable through UV detection. However, mass confirmation of the analyte requires mass spectrometry. Additionally, quality control necessitates fast and robust methods to ensure high throughput. These can be achieved by combining instruments such as the Thermo Scientific ISQ EM single quadrupole mass spectrometer and the Thermo Scientific Vanquish Flex binary UHPLC system. This paper introduces two workflows for data analysis. Both use peak areas from UV and MS detection to determine the relative abundance of analytes within the sample. However, the first workflow identifies analytes based on the ion species with the highest signal intensity at m/z. The second workflow employs deconvolution of mass spectra for mass determination and analyte identification.

Evaluation of pH Shift in Glass Vials for Auto-Samplers

Do glass vials affect the pH of valuable samples? The answer is simple: "Yes."

pH has a significant impact on the ionization state of specific analytes. For example, a base with a pKa of 8.5 is 99% ionized at pH 6.5 using the 2 pH unit rule and 99% non-ionized at pH 10.5. When the analyte is close to its own pKa, unstable chromatograms may be observed, so it is necessary to adjust the pH to achieve the appropriate ionization state. Differences in the surface activity of vial glass and the leaching of hydrolysis products that affect the sample pH can be confirmed by monitoring the pH changes of deionized water stored in the vial. To investigate how the quality of glass, represented by the expansion coefficient, affects the pH of the solution, we measured the fluctuations in the pH of 18 MΩ pure water stored in an autosampler vial for a certain period at room temperature and compared the results with those obtained using polypropylene containers as a control.
