We synthesize RNA through in vitro transcription using T7 polymerase from template DNA (prepared from plasmid clones via PCR or oligo DNA), followed by purification with silica columns. The purity and yield are confirmed through absorbance measurement and electrophoresis. In "ssODN (single-stranded oligodeoxynucleotide donor DNA) synthesis," it can be used as donor DNA to knock in sequences of up to several dozen bases, including base substitutions and loxP sites.
