Investigating the effects of compounds on full-length kinase proteins expressed in intact cells.
To more accurately predict the efficacy of drugs against kinases, it is extremely important to evaluate the interactions between target proteins and compounds in a cellular environment that closely resembles in vivo conditions, and to quantitatively measure the drug's inhibitory ability, selectivity, and affinity. In addition to measurements at equilibrium, it is also desirable to assess kinetics and consider kinetic parameters such as the drug's residence time in the optimization of drugs. Our cell-based assay service, using the NanoBRET(TM) Target Engagement (TE) Intracellular Kinase Assay System (from Promega), allows you to select your desired kinase target from a list and evaluate how your compound interacts with that target under conditions close to in vivo, providing you with quantitative results. By simply sending us your compound, you can obtain IC50 values, inhibition rates, and residence time evaluation results measured in intact cells.

This is an assay for detecting protein interactions in living cells based on energy transfer from the donor, NanoLuc(R) luciferase fusion protein, to the fluorescently labeled probe, NanoBRET(TM) tracer. By using compounds that competitively replace the NanoBRET(TM) tracer binding to the NanoLuc(R) luciferase fusion protein expressed in living cells, the apparent affinity of the compounds can be measured through reversible binding. In addition to measuring inhibition rates and IC50 values, we also offer a Residence Time analysis service that measures the residence time of drugs on target proteins over time and analyzes their characteristics.
