Genovis AB IgGZERO (EndoS)

Endoglycosidases useful for the analysis and evaluation of antibody drugs and ADCs (antibody-drug conjugates) including biosimilars.

IgGZERO (EndoS) is an endoglycosidase derived from Streptococcus pyogenes that hydrolyzes the N-linked glycans in the Fc region of IgG. It is useful for reducing complexity through rapid deglycosylation of IgG and inactivating the effector functions of the Fc region. This enzyme deglycosylates IgG from various animal species, including human IgG (all subclasses), as well as IgG from mice, rats, monkeys, sheep, goats, cows, and horses. The enzyme specifically hydrolyzes only the glycans in the Fc region of IgG, even in the presence of other glycosylated proteins in solution. It is expressed and produced in E. coli, contains a His tag, and has a molecular weight of 110 kDa. Genovis actively holds web meetings and seminars. Introducing Genovis enzymes useful for the evaluation and analysis of antibody-drug conjugates. ◎FabRICATOR (IdeS) An enzyme that digests specific sites just below the hinge region of antibodies. It generates a uniform pool of F(ab')2 and Fc/2 with a 30-minute incubation. Typically, IgG is digested within 30 minutes, but there is no risk of excessive digestion even with longer incubation times. The enzyme activity is maximized at 37°C, but it can also be used at room temperature by extending the reaction time. ◎IgGZERO (EndoS) An enzyme that hydrolyzes the N-linked glycans in the Fc region of IgG. It is useful for reducing complexity through rapid deglycosylation of IgG and inactivating the effector functions of the Fc region. ◎GlycINATOR (EndoS2) An enzyme that hydrolyzes the glycans in the Fc region of IgG. Genovis Inc. Immobilized PNGase F

Rapid and easy removal of N-glycans from antibodies and proteins using a spin column.

Immobilized PNGase F is a product in which the enzyme PNGase F (Peptide N-glycosidase F) is covalently bonded to agarose beads in a spin column format, allowing for the easy removal of N-glycans from antibodies, fusion proteins, and other N-glycosylated proteins. This enzyme is produced by expressing a gene derived from Elizabethkingia meningoseptica in E. coli through recombinant methods. Samples of glycoproteins are incubated with Immobilized PNGase F under standard reaction conditions for 1 hour to overnight, or under denaturing reaction conditions (using RapiGest SF Surfactant) for 15 to 60 minutes. The deglycosylated glycoproteins can then be easily collected by centrifugation. The activity of PNGase F on some glycoproteins may be reduced or inhibited due to steric hindrance. In such cases, longer incubation times or denaturation of the glycoproteins may be necessary.
