World's First Holographic Fluorescence Microscope for Cell Observation

The 3D Cell Explorer-fluo, a holographic and fluorescence microscope for cell observation, is an innovative device that combines 3D refractive index imaging and 2D fluorescence imaging using holography and tomography technologies. It allows for rapid and easy observation of the internal structures and organelle distributions of cells and tissues. By overlaying fluorescence images of various cellular organelles and cell membranes that have been fluorescently labeled with refractive index images, it is possible to calibrate the digital staining to match the staining images of the fluorescent labels. By applying this calibrated digital staining to the 3D refractive index image, the 2D fluorescence images can be extended into 3D images, visualizing the three-dimensional distribution. This calibration process allows for a correlation between fluorescence staining and digital staining, enabling compatibility with unstained samples. The constructed 3D images can be enlarged, reduced, rotated, and displayed in cross-sections, allowing for comprehensive observation of the interior of cells from various perspectives.
