High-sensitivity immunoassay using nanoantibody-presenting polysaccharides [Saitama University]
By combining nanoantibodies with polysaccharides to create "nanoantibody-presenting polysaccharides," it is possible to immobilize nanoantibodies onto substrates.
○Overview of the Technology We have developed a method to improve detection sensitivity using enzyme-linked immunosorbent assay (ELISA) with nanobodies. We prepared a nanobody-polysaccharide conjugate by chemically linking nanobodies to water-soluble polysaccharides, and it was confirmed that the sensitivity in the solid-phase ELISA using this conjugate was improved compared to using the nanobodies alone. ○Comparison with Conventional Technology In ELISA using nanobodies as a substitute for conventional antibodies, there was a challenge of inactivation due to solid-phase immobilization and detachment from the substrate. This technology allows for the improvement of ELISA detection sensitivity by using water-soluble polysaccharides as a scaffold for solid-phase immobilization of nanobodies. ○Features of the Technology - Application of nanobodies in sandwich ELISA - Water-soluble polysaccharides protect nanobodies - Improved binding affinity due to aggregation of nanobodies - Suppression of nonspecific adsorption
