By controlling the concentration of nitrites in drugs and processes, the formation of nitrosamines can be prevented, making it essential to have a highly sensitive method for analyzing nitrites in pharmaceuticals and their raw materials. In many cases, dimethylamine is used in the synthesis of various drugs. Under acidic conditions, it reacts with nitrites to form nitrosamines. This also applies to the production of hydroxypropyl methylcellulose (HPMC), a common excipient. This technical document introduces the quantification of nitrites in hydroxypropyl methylcellulose using ion chromatography (IC) with direct UV/VIS detection.

Zinc oxide is a powdered mineral that adheres to the outermost layer of the skin, acting as a broad-spectrum blocking agent that scatters, reflects, and absorbs ultraviolet radiation, providing protection from both UVA and UVB radiation, making it unique among sunscreen ingredients. The United States Pharmacopeia (USP) has developed monographs for FDA-approved drugs, including OTC medications, and is working to obtain the necessary information to maintain high-quality pharmaceuticals. It has also initiated a global effort to update many existing monographs across all official methods. As part of this USP initiative, the addition of an ion chromatography (IC) method for zinc measurement in General Chapter <591> is proposed. Gradient analysis is a method that enhances separation by increasing the concentration of the eluent from low to high during the analysis, allowing components that are retained more easily to elute faster. We will introduce examples of the separation of inorganic anions and organic acids regarding the separation of ionic species due to changes in eluent concentration and the setting of gradient conditions. By using an eluent generator system, it is possible to supply accurate and high-purity eluent simply by sending ultrapure water. Additionally, gradient analysis can be easily performed with just the concentration settings from the workstation.
