In general, plasmid DNA is obtained through purification from E. coli culture broth. The culture broth contains not only host-derived genomic DNA, RNA, and proteins, which are impurities, but also open circular DNA (Open circular) and linear DNA, which are structural isomers of the target plasmid. These can reduce the efficiency of gene expression, so clinical plasmid DNA is required to have a purity of over 90% for supercoiled DNA. Therefore, in this study, we investigated the separation of structural isomers of plasmid DNA pBR322 using anion exchange chromatography, and we would like to introduce it.
