The chromatogram shows the separation and detection of anthracene and pyrene by HPLC (high-performance liquid chromatography).
The only difference in the mobile phase used is whether it has been degassed using a degasser (R) or not; all other conditions, including sample concentration and sample volume, are exactly the same. However, even with just the difference in degassing conditions, the peak size of pyrene shows completely different results.

basic information
【Features】 ○ Fluorescence detection involves the sample emitting fluorescence when illuminated by light, and quantifying trace components by measuring that fluorescence. ○ It has long been recognized that dissolved oxygen in liquids can interfere with fluorescence emission or absorb fluorescence after emission, leading to a decrease in fluorescence intensity; thus, the differences observed in this HPLC analysis are to be expected. ○ An investigation into the relationship between the dissolved oxygen concentration in the mobile phase and the intensity of the pyrene peak revealed that the peak intensity increases sharply when the dissolved oxygen concentration is below 3 ppm. ○ Of course, these changes vary depending on the sample material and wavelength conditions (excitation light wavelength, fluorescence wavelength), and the degree of influence from dissolved oxygen also differs. ○ This research report has been presented at domestic and international conferences, resulting in a correction of the perception that mobile phase degassing in HPLC is merely for preventing foaming. ○ It is now considered that degassing is an essential technique for ensuring the accuracy of analytical measurements of various liquids. ● For more details, please contact us or refer to the catalog.
