CHO Cell Culture Optimization: A DOE Approach in the Ambr 15

Selecting the appropriate combination of culture media and feeds for cells, and further optimizing process parameters, is a complex and time-consuming process. Sartorius's extensive portfolio of media and micro-bioreactors that allow for parallel cultivation support optimal and scalable process development for cells. This application note introduces the process optimization of CHO cell culture using Sartorius media, Ambr15, and the experimental design software MODDE.

Our research has confirmed the following: - Maintenance of stem cell markers and retention of differentiation potential - No enlargement of cell diameter or aging phenomena - No chromosomal changes that could lead to tumorigenesis, and no tumor formation after transplantation ■Cell Recovery Rate and Viability Over 90% ■Adjustment of Cultured Cell Numbers It is possible to adjust the number of cells by modifying the amount of non-woven fabric that serves as the cell scaffold.
