Fully Automatic Western "Jess"

The catabolism of branched-chain amino acids (BCAA) is important for the energy metabolism of the whole body, including skeletal muscle, and abnormalities in this process have been noted to be associated with various metabolic diseases. The BCKDH complex, which is responsible for the rate-limiting step of BCAA catabolism, is regulated by the reversible phosphorylation of the E1α subunit by BDK and PPM1K. In this seminar, using skeletal muscle cells, we will present research showing that lauric acid suppresses BCAA oxidation through an O-GlcNAc-sensitive BCKDH regulatory mechanism, and we will introduce the use of a fully automated capillary western system for the analysis of BCKDH phosphorylation and global O-GlcNAcylation.

In liquid biopsy research, we will introduce a technical evaluation of proteome analysis to obtain reliable data from trace plasma, as well as case studies on data quality verification using the Contamination Index and Simple Western.

We are utilizing Jess in the presentation paper. We will hold an online seminar featuring a presentation by Kiyoka Deguchi, Lecturer at the Institute of Intractable Diseases, Graduate School of Science and Technology, Tokyo University of Science. The lecture by Professor Ryusuke Niwa from the Center for Survival Dynamics Research at Tsukuba University, focusing on physiological genetics research, is now available for viewing.
