HexAuFoil samples support improves every image! CryoEM inherently has issues with data loss caused by particle movement during imaging, which leads to a decrease in mapping quality. While data processing can mitigate this data loss, advanced sample processing techniques are required. To address these issues, we propose a solution using the new cryoEM grid "HexAuFoil," which has been launched by Quantifoil, a group company of SPT Labtech.

We assist in the creation of high-quality Cryo-EM samples. Thanks to the nanowires on the grid surface, blotting of the samples is unnecessary. ● High-speed sample processing without blotting Using hygroscopic grids treated with nanowires allows for rapid sample dispensing in the air. This reduces the effects of the gas-liquid interface and may address issues related to preferred orientation, aggregation, or denaturation. ● Automated grid processing A series of operations from sample application to freezing of the grid set in the grid holder is automated. The mosquito LCP is a nanoliter dispensing machine for membrane protein crystallization, equipped with a dedicated dispensing head for the LCP crystallization method, in addition to the functions of the Xtal3 model. It allows for the miniaturization of rare protein sample amounts and enables reproducible high-throughput experiments. We also offer a precision humidification chamber (optional) to mitigate the effects of drying in miniaturized experiments. This is a mosquito-specific chamber that controls the humidity around the dispensing deck. The LCP model comes in two types: a 2-way type that can accommodate two crystallization plates and a 4-way type that can accommodate four plates. Dragonfly Crystal (for protein crystallization) This is a new non-contact dispenser that employs both positive displacement technology and disposable syringes. It allows for the rapid dispensing of crystallization buffers prepared from multiple components directly onto crystallization plates with low dead volume, and can seamlessly transition to the mosquito. It is also convenient for dispensing Master Mix and other solutions into PCR plates. ● Positive Displacement Technology With a unique non-contact dispensing method, it dispenses various viscous solutions with high precision. Since it uses disposable tips, there is no need for troublesome cleaning, eliminating the risk of contamination and clogging. ● User-Friendly Dedicated Software The dedicated software allows for easy setup of conditions such as combinations of precipitants, buffers, salts, concentration gradients, and pH gradients, all through intuitive operation.

The mosquito Xtal3 is a versatile and robust nanoliter-scale dispensing device that employs a positive displacement method. It allows for the miniaturization of rare protein sample volumes and enables reproducible high-throughput experiments. In addition to precise aspiration and dispensing, it can also perform accurate mixing, accommodating various applications in protein crystallization.
