Performing G protein-coupled receptor (GPCR) assays using live cells in a high-throughput manner is an important part of the drug discovery process for small molecules. By combining the Fluent laboratory automation solution designed for live cell assays with the DiscoveRx PathHunter β-Arrestin human and ortholog GPCR assay kits, 384-well plate-based assays can be executed in high throughput. The NCI-60 panel is a collection of various cancer cell lines and is one of the most important tools in drug discovery for evaluating the activity of anticancer agents. After sample preparation using Fluent, cell viability was measured using luciferase detection for cellular ATP measurement (CellTiter-Glo, Promega).
