Comparison of the performance of our currently used serum-containing medium and iMediam for T (Day 14 of culture) Peripheral blood mononuclear cells (PBMCs) were isolated from the blood of 9 healthy individuals who provided informed consent, and a comparative culture was conducted using our serum-containing medium and iMediam for T (serum-free). Cultures were maintained in OKT3 antibody-coated flasks for the first 5 days, after which they were transferred to gas-permeable culture bags, resulting in a total culture period of 14 days. The final liquid volume was set at 2,000 mL.

Surface antigen analysis After culturing PBMCs from three healthy individuals who provided informed consent in iMediam for T (serum-free) for 14 days, the proportion of CD3-positive cells, which are indicative of T lymphocytes, was measured using a flow cytometer. In all three samples, the proportion of CD3 was over 98%, indicating that T lymphocytes are selectively proliferating.
