【Sample Analysis】Nine types of whiskey with different brands (A to I) 【Analysis Method】The diluted samples were subjected to dynamic headspace (DHS) - GC/MS analysis. After deconvoluting the peaks obtained from the chromatogram (GC/MS-TIC), the aroma components were identified using an automated comprehensive search of the aroma component database (Aroma Search). For the identified aroma components, multivariate analysis (principal component analysis) was conducted on those with variations within the specified range over three repeated measurements. 【Analysis Results】The results of the multivariate analysis (principal component analysis) are shown in a score plot indicating the differences between samples and a loading plot indicating the components responsible for these differences. The score plot revealed that the method effectively distinguishes between types of whiskey. The loading plot confirmed the presence of phenolic compounds characteristic of Islay whiskey, which has a complex and deep aroma derived from peat.

We introduce examples of significant contributions to FANTOM, which has developed and expanded around the field of transcriptome analysis. At FANTOM, we analyzed the expression profiles of mature miRNAs and primary transcripts in numerous primary cultured cells using short RNA-seq analysis and the CAGE method. Furthermore, the CAGE method revealed these promoter sequences. In the conventional "CAGE" analysis, total RNA is input, allowing for the following: - RNA expression analysis (serving the same role as general RNA-seq) - Identification of transcription start sites (the greatest strength of CAGE, which is insufficient with RNA-seq) - Searching for transcription factor binding motifs based on the identified transcription start site data (unique to CAGE, a reverse search function of ChIP-seq) The central principle of library preparation is the "cap trap method," a unique technology developed by RIKEN that captures the cap structure at the 5' end of RNA. In this "NET-CAGE" analysis, "NET-RNA," which is currently being synthesized in the nucleus, is extracted and recovered using a unique technique and input for library preparation. (NET-RNA: Negative Elongating-Transcript: newly synthesized RNA) Generally, the lifespan of total RNA is relatively long, ranging from several hours to several days. In contrast, the eRNA (enhancer RNA) from NET-CAGE has an extremely short lifespan, being degraded within just a few minutes. "CAGE" allows us to observe the total RNA present in the cell, while "NET-CAGE" enables us to see the NET-RNA that is currently being synthesized. It has become possible to differentiate between the points we want to observe.
