By applying this calibrated digital staining to the 3D refractive index image, the 2D fluorescence images can be extended into 3D images, visualizing the three-dimensional distribution. This calibration process allows for a correlation between fluorescence staining and digital staining, enabling compatibility with unstained samples. The constructed 3D images can be enlarged, reduced, rotated, and displayed in cross-sections, allowing for comprehensive observation of the interior of cells from various perspectives.
