Utilizing the features of a unique antibody production technology that does not rely on animals! An article introducing strategic selection.

The monoclonal antibody production service by Antibody Craftsman utilizes a unique antibody production technology that does not rely on animals, allowing for in vitro selection to narrow down the regions of antigens that antibodies bind to and to create antibodies that can distinguish slight differences in antigens. Here, we introduce some of the selection methods used to obtain the desired Fab antibodies. Additionally, please take a look at our achievements in antibody production. [Content Included] ■ Antibodies that identify post-translational modifications ■ Antibodies that specifically recognize homologous regions ■ Antibodies against membrane proteins ■ Antibodies that distinguish amino acid substitutions and deletions ■ Antibodies that recognize three-dimensional structures ■ Antibodies with strict specificity *For detailed content, please refer to the related links. *For detailed specifications, please refer to the PDF materials or feel free to contact us.

Deliberately selecting antibodies that are recognized by autotaxin! An article introducing examples of antibodies that inhibit activity.

We will introduce a case of obtaining antibodies that inhibit activity, which could not be achieved through animal immunization, using the phage display method called "Antibody Craftsman," specifically in the case of autotaxin. The reason why inhibitory antibodies against autotaxin (ATX) could not be obtained through animal immunization is thought to be due to the high conservation of ATX across species, making it difficult to establish inhibitory antibodies targeting the catalytic active site. Therefore, in the Antibody Craftsman approach, we intentionally selected antibodies that recognize both human and mouse autotaxin (ATX) and developed a strategy to select inhibitory antibodies from among them. 【Contents】 1. What is Autotaxin 2. Structure of Autotaxin 3. Strategic Screening - 3-1. Panning - 3-2. ELSIA Screening - 3-3. Enzyme Activity Inhibition Screening 4. Activity Measurement Using Antibodies 5. Summary *For detailed content of the article, please refer to the related links. *For detailed specifications, please refer to the PDF materials or feel free to contact us.

Selection of antibodies that bind only to mutant types! An article introducing the production of antibodies that distinguish between mutant types differing by just one amino acid.

In cases where a protein with a single amino acid mutation exists in a mixed state alongside the wild-type sequence, there may be a need for antibodies that can specifically distinguish this mutated protein. Specifically, as an antigen, a peptide synthesized from the mutated sequence portion is used, and through panning via phage display and ELISA screening, clones that are negative for the wild-type and positive for the peptide with the single amino acid mutation are selected. I will introduce each step. [Contents] 1. Design and coupling of peptide antigens 2. Panning 3. ELISA screening 4. Sequencing 5. Selection of candidate antibodies *For detailed content of the article, please refer to the related links. *For detailed specifications, please refer to the PDF materials or feel free to contact us.

Setting the antigens is very important! An article introducing the selection of antibodies that distinguish the differences at the terminal ends.

When proteins undergo processing and are divided into multiple fragments, it becomes very important to set the antigens for producing antibodies that recognize each fragment. Antibody artisans perform all selections in vitro without using animal immunization, allowing them to conduct binding reactions with peptides or proteins in their prepared state, without the steps of fragmenting proteins for antigen presentation. In this article, we will introduce the antigen setting and selection methods for producing antibodies that can distinguish between fragments before and after cleavage by a protease of a certain protein. 【Contents】 ■1. Specificity of antibodies ■2. Design of antigens ■3. Screening methods ・3-1. Panning ・3-2. ELISA screening ■4. Protein antigens and peptide antigens ・4-1. Which one should be chosen? ・4-2. Innovations in selection ■5. Summary *For detailed content of the article, please refer to the related links. *For detailed specifications, please refer to the PDF materials or feel free to contact us.

It is possible to narrow down the field! An article introducing antibodies that distinguish phosphorylated Ser.

Antibody artisans conduct selection in vitro against fixed antigens without using animal immunity. This allows for binding reactions to be performed in the state or environment of prepared peptides or proteins, enabling precise antigen design, such as narrowing down the area of interest. Here, we present an example of antibody production using peptide antigens to obtain antibodies that recognize specifically phosphorylated proteins at certain Ser residues. We invite you to read on. [Contents] 1. Preparation of Antigens 2. Selection to Increase Specificity 3. Selection of Antibodies 4. Summary *For detailed content of the article, please refer to the related links. *For detailed specifications, please refer to the PDF materials or feel free to contact us.

Monoclonal Antibody Production Contract Service "Antibody Craftsman"

Success-based reward! We will deliver antibodies that distinguish slight differences in 7 weeks.

Are you having trouble with the low specificity of conventional monoclonal antibodies? With "Antibody Artisan," you can produce antibodies targeting specific regions, resulting in antibodies with higher specificity. For example, we can produce antibodies such as: ■ Antibodies that identify post-translational modifications ■ Antibodies that specifically recognize homologous regions ■ Antibodies that distinguish between a few amino acid substitutions or deletions ■ Antibodies that recognize denatured proteins "Antibody Artisan" employs a unique technology (a library of 45 billion types of artificial human antibodies and phage display) that does not use animal immunization, allowing us to design antigens and select strategically. All work is done in vitro, enabling us to deliver antibodies as quickly as seven weeks after receiving the antigen. Additionally, the antibodies obtained through this service can be expressed and purified as recombinant proteins in E. coli, allowing for repeated production of additional antibodies. Antibody clones are stored for five years, and sequences are kept permanently. *For more details, please refer to the PDF materials or feel free to contact us.

Ibeck Corporation Monoclonal Antibody Production Service

We will assist in the practical application of drug discovery seeds and the development of pharmaceutical candidate antibodies.

We use human PBMCs (peripheral blood lymphocytes) as materials to clone the target antibody gene using the EBV method and produce fully human monoclonal antibodies. We create a library of healthy human samples consisting of over 100 specimens to assess the likelihood of obtaining the target antibody from healthy individuals. If there are specimens from which we can determine a high probability of acquiring the target antibody based on the results obtained, we can start antibody screening using our owned sample specimens. We emphasize acquiring antibodies that maintain their original activity, producing full-length heavy and light chains without converting them to scFv. 【Features】 - Application of the EBV method - Cloning of the target antibody gene and production of fully human monoclonal antibodies - Emphasis on acquiring antibodies that maintain their original activity - No scFv conversion *For more details, please refer to the catalog or feel free to contact us.

Monoclonal Antibody Production Contract Service: Technical Information 2

Unique Antibody Production Technology - HuCAL Technology

HuCAL technology is an antibody production technique that does not rely on animals and conducts all selections in vitro. Specifically, antibodies are presented using phages that incorporate human antibody genes, allowing them to bind to antigens. Since the presented antibodies (Fab antibodies) correspond one-to-one with the genes that code for them in the form of phage particles, it is easy to identify the genes of the Fab antibodies that bind to the antigen from among many Fab antibodies (phage display). A solution containing phages is added to a plate with immobilized antigens, allowing the phages to bind through the presented antibodies. Phages that did not bind to the antigens are washed away, and the bound phages are then eluted and collected. By repeating this process several times, phages that bind more strongly to the antigen can be concentrated (panning). The antibody genes encoded by the phages obtained through panning are expressed in E. coli, and Fab antibodies are purified. *For more details, please refer to the PDF materials or feel free to contact us.*
