Many life phenomena, including various diseases, are caused by biomolecules such as proteins, and observing molecular structures and dynamics in the microscopic realm is crucial for understanding these phenomena and for fundamental treatments of diseases. Molecular dynamics simulation is a technique that allows for the "direct" observation of microscopic behavior by reconstructing biomolecular models with atomic resolution in a computer and moving the molecules according to physical laws. Coupled with the computational power of computers, this technique has developed to the point where it is referred to as a computational microscope and is actively used as a method to complement experiments. However, there are two challenges to contributing to drug discovery and materials development: "too much computation time" and "limitations in model accuracy." To address the first challenge, we are working on introducing efficient algorithms to predict loop structures of next-generation antibodies in a shorter time. For the second challenge, we are developing methods that integrate experimental data and simulations using statistical mathematics and machine learning to achieve more accurate observations.

One method to observe neuronal activity during free behavior is the head-mounted fluorescence microscope.

One method for observing neuronal activity under free behavior is the head-mounted fluorescence microscope. The microscope body weighs only 3 grams and is equipped with a CMOS sensor, an electronic focusing mechanism (focal depth 0-300μm), and filters. An imaging cannula (GRIN lens) is inserted into the animal's head, capturing images at the lens tip with the CMOS sensor. This model adds a light stimulation function (optogenetics): NpHR halorhodopsin activation capability to the aforementioned features. The microscope body and the imaging cannula (GRIN lens) are securely and easily fixed with screws, and no special fixtures are required for fixation. Four types of imaging cannulas are available, depending on the depth of the target to be observed and the diameter of the GRIN lens. By using an imaging cannula equipped with a prism at the tip, it is also possible to observe sagittal or coronal sections.

The photos showing the structural color of the sample were taken with a digital camera and a digital microscope (KEYENCE VHX-500). For angle dependence, two types of illumination were used. The first type (diffused light) is illumination that does not come from a specific direction, and the sample is illuminated by several ceiling lights and secondary scattering from the walls. The second type is illumination via fiber (Olympus, LG-PS2). The illumination light is directed from approximately 50 degrees tilted from the normal direction.
